A Study on the Lactic Dehydrogenase Isoenzyme of the Rabbit Kidney

Abstract
"Rabbit kidneys were perfused, separated into the cortex and medulla, and subjected to cellulose acetate electrophoresis to observe their LDH isoenzyme patterns. The effect of acetone on the patterns was also studied on both tissues. H4_3nd M4_LDH were partially purified from the cardiac and skeletal muscles of the rabbit, respectively, and their in vitro hybridation was carried out. The following conclusions were made 1) In the rabbit kidney, activity of LDH-I is higher in the cortex, while those of LDH-4 and-5 are higher in the medulla, disclosing that the distribution patterns of the LDH isoenzyme are different between the two tissues of the rabbit kidney 2) High concentration of acetone inactivates M.LDH of the cortex and medulla selectively, leaving H_LDH of both tissues unaffected, which might provide an evidence to differentiate between the cortex and medullary diseases. 3) It is po3sible, with the use of DEAE-cellulose, to obtain more than five-fold partially purified H4-3nd k1.-LDH from the rabbit cardiac and skeletal muscles, respectively. The two preparations can undergo hybridation in vitro however, it is not likely to reproduce in vivo LDH isoenzyme pattern of the rabbit kidney under the given condition."
